==== Dangers ==== If taken undiluted by mouth, GBL can cause esophageal and gastro-intestinal irritation. It is possible for oral ingestion of GBL to cause nausea and other similar problems, possibly more so than with GHB. GHB has biphasic effects, a euphoric effect at low doses (the reason for the term liquid ecstasy), and a sedative effect at higher doses. As a result of this sedation it can cause unconsciousness. When combined with alcohol the increased sedation and risk of vomiting results in a high risk of fatality. As a result, many harm reduction organizations suggest never mixing the two drugs. There have been several news reports of deaths associated with GBL, usually in combination with alcohol or other depressants.

When the Soviets arrived, Prague was already in a general state of confusion due to the Prague Uprising. Soviet and other Allied troops were withdrawn from Czechoslovakia in the same year. ==== DNA replication ==== DNA replication has been shown to affect the function of various DNA repair enzymes. H-DNA formation involves the formation of single-stranded DNA (ssDNA), which is more susceptible to attack by nucleases. Various nucleases have been shown to interact with H-DNA in a replication-dependent or replication-independent manner. A study using human cells found that the nucleotide excision repair (NER) nucleases ERCC1-XPF and ERCC1-XPG induced genetic instability. These enzymes cleave H-DNA at the loop formed by the two Hoogsteen hydrogen-bonded strands and the 5' end of the other Watson-Crick hydrogen-bonded strand, respectively. This cleavage has been shown to induce large deletions that cause double strand breaks (DSBs) in DNA that can lead to genetic instability. In cells deficient in ERCC1-XPF and ERCC1-XPG, these deletions were less prevalent near H-DNA forming sequences. Additionally, more mutations were found in ERCC1-XPF and ERCC1-XPG deficient cells in the absence of DNA replication, which suggests they process H-DNA in a replication-independent manner. Alternatively, the DNA-replication repair nuclease FEN1 was found to suppress genetic instability. Similar to ERCC1-XPG, FEN1 cleaves H-DNA at the 5' end of the strand not involved in Hoogsteen hydrogen-bonding. HeLa cells deficient in FEN1 showed higher prevalence of deletions near H-DNA forming sequences, but H-DNA induced mutagenesis was more pronounced in FEN1 deficient cells in the presence of DNA replication.

==== Absorption and bioavailability ==== The oral bioavailability of estradiol is very low. This is due to the fact that estradiol is poorly soluble in water, which limits its dissolution and absorption, and is additionally subject to extensive metabolism during the first pass through the intestines and liver. Estradiol is micronized and/or conjugated with an ester, as in estradiol valerate or estradiol acetate, to improve its oral bioavailability and potency. Micronization decreases the particle size of estradiol crystals and hence increases the surface area for absorption, thereby improving the rate and extent of absorption. In addition, there is an improvement in metabolic stability. Oral micronized estradiol consists of more than 80% of estradiol particles micronized to a size smaller than 20 μm in diameter, or to about 1 to 3 μm on average. All oral formulations of estradiol available today are micronized, and oral estradiol valerate tablets also seem to be micronized. Oral non-micronized estradiol and oral micronized estradiol do not appear to have ever been directly compared in a study. Both have been assessed independently however, and have been found to produce significant estrogenic effects. Micronization of other poorly water-soluble steroids such as spironolactone and norethisterone acetate has been found to increase their potency by several-fold.
