BDNF raises a handful of sensible questions. This page answers them in order, starting with the fundamentals and moving to applications.
Reviewed 2025-09-04. Anything still debated is marked as such rather than presented as settled.
Published clinical work is concentrated in Russian-language journals and generally involves small samples without independent replication. Systematic reviews in English note the shortage of randomised, placebo-controlled trials and the difficulty of verifying methods from translated reports. Outcome measures vary between studies, which complicates pooling of results. Interest in the compound as a cognitive or anxiolytic agent therefore rests on a thinner evidence base than the volume of citations suggests. Replication in well-powered trials with preregistered endpoints would be needed before firm conclusions about efficacy can be drawn.
Proposed mechanisms centre on the GABAergic system. Animal and tissue studies report changes in GABA-A receptor expression and reduced activity of GABA transaminase, the enzyme that degrades GABA. Effects on monoamine turnover, including serotonin and dopamine pathways, are also described, and a separate line of work links the peptide to increased expression of brain-derived neurotrophic factor in hippocampal tissue. Most of these findings come from rodent models and cell preparations. How the individual observations combine into a single coherent mode of action is not settled.
Pharmacokinetic data are sparse and largely derived from animal work. After intranasal administration the peptide appears in plasma within minutes, and reported half-lives are short, on the order of minutes to tens of minutes. Degradation proceeds through ordinary proteolytic cleavage into constituent amino acids and smaller fragments. Direct evidence that intact Selank reaches brain tissue in meaningful amounts is limited, and the extent of blood-brain barrier penetration is debated. Some authors argue that fragments, not the parent peptide, carry much of the observed activity.
Measuring peptide exposure inside the brain is technically difficult. Selank is degraded rapidly in plasma, and assays must separate intact peptide from fragments, which favors targeted mass spectrometry over immunoassays alone. Reported half-lives are short, on the order of minutes, so effects observed hours later are attributed to downstream signaling rather than to the parent compound. Blood-brain barrier permeability is debated and rarely quantified directly. Gaps include absent dose-response characterization, inconsistent reporting of purity, and almost no pharmacokinetic data from human participants.
Selank is studied chiefly as an animal-model anxiolytic with proposed secondary effects on memory and immune signaling. Reported mechanisms include modulation of the GABA-A receptor complex, inhibition of enkephalin-degrading enzymes, and shifts in monoamine turnover within limbic structures. Some experiments describe increased expression of brain-derived neurotrophic factor in the hippocampus after repeated dosing. No single molecular target has been confirmed, and the peptide does not bind any receptor with the selectivity typical of a conventional small-molecule drug. Mechanism therefore remains a set of hypotheses rather than an established pathway.
| Property | Value | Notes |
|---|---|---|
| Primary route studied | Intranasal | Also examined parenterally in animal work |
| Reported plasma half-life | Minutes to tens of minutes | Values vary widely between reports |
| Main model systems | Rodent behavioural and cell assays | Human trials are few and small |
| Principal proposed targets | GABA-A receptor, GABA transaminase | Monoamine and neurotrophic pathways also reported |
| Evidence grade | Preliminary | Limited independent replication |
Animal studies have examined behaviour in tests of anxiety, memory retention and stress response, and several report changes in neurotrophic or neurotransmitter-related markers. The human evidence base is much smaller, consisting mainly of short trials conducted in Russia with limited reporting in English-language journals. Sample sizes are modest and outcome measures vary between studies, so the findings are best described as preliminary. Independent replication under modern trial standards has not been widely reported.
Outside its country of origin the compound is generally handled as a research chemical rather than an approved medicine. No regulatory approval from the United States Food and Drug Administration or the European Medicines Agency has been granted for human use. Identity and purity are normally checked by reverse-phase high-performance liquid chromatography, with mass spectrometry used to confirm the molecular mass. Lyophilised material is stored cold and desiccated, and repeated freeze-thaw cycles are avoided.
Published work on this peptide almost always uses intranasal delivery, with drops or a spray applied to the nasal mucosa. Some animal experiments have used subcutaneous or intraperitoneal injection, and a smaller number have compared routes directly. Oral administration is not a focus of the literature, because short peptides of this size are broken down by digestive enzymes and cross intestinal barriers poorly. How much of an intranasal dose reaches the bloodstream intact in humans remains an open question.
Enzymatic stability motivates the extra three residues at the carboxyl end. Native tuftsin is cleaved quickly by circulating aminopeptidases and carboxypeptidases, which limits its duration of action and its usefulness as a tool compound. Extending the chain with proline-rich segments is a common design tactic because proline constrains the backbone and slows proteolysis. The same Pro-Gly-Pro motif appears in other Russian-developed peptides of the era. Whether the full seven-residue chain is required for activity, or whether it acts mainly as a prodrug releasing tuftsin, remains unresolved.
Selank is a synthetic heptapeptide with the sequence Thr-Lys-Pro-Arg-Pro-Gly-Pro. Its four N-terminal residues reproduce tuftsin, a tetrapeptide fragment of the immunoglobulin heavy chain, while the C-terminal Pro-Gly-Pro extension is a synthetic addition. The peptide has a molecular mass near 752 daltons and carries a net positive charge at physiological pH because of the arginine and lysine side chains. Published indexes list it under the name Selank and the sequence abbreviation TKPRPGP. Solid-phase peptide synthesis is the standard production route for research quantities.
Experts said that their findings did not challenge the conclusions by UK government: "We provided that information to the Government who have then used a number of other sources to come to the conclusions that they have." On 12 April 2018 the OPCW announced that their investigations agreed with the conclusions made by the UK about the identity of the chemical used. By September 2018, two Russian "tourists", "Alexander Petrov" and "Ruslan Boshirov", had been identified as suspects. They told Margarita Simonyan, the chief editor of RT television, in an interview that they both worked in the sports nutrition business and that: "Those are our real names.. We're afraid to go out, we fear for ourselves, our lives and lives of our loved ones." The Crown Prosecution Service announced enough evidence was obtained by that date "to convict the two men" of the attack, although it did not apply to Russia "for their extradition because Russia does not extradite its own nationals. [...] However, a European Arrest Warrant has been obtained in case they travel to the EU". In February 2019, the Bellingcat website published precise allegations that identified GRU Major Denis Vyacheslavovich Sergeev as a man who travelled in March 2018 to London under the false identity of Sergei Fedotov. It is claimed with detailed photograph evidence, and phone, travel, passport, and motoring database records that GRU Colonels Alexander Mishkin and Anatoly Chepiga assumed the identities of Petrov and Boshirov, and placed the poison on Skripal's doorknob.
=== Political involvements === In 2022, Tiu Laurel donated ₱30 million to the Partido Federal ng Pilipinas for the presidential campaign of its nominee Bongbong Marcos, establishing himself as the top donor. Additionally, he made contributions of ₱20 million and nearly ₱8.75 million for the successful senatorial campaigns of Migz Zubiri and Robin Padilla, respectively.
The Bundeswehr uses the Einpersonenpackung to provide two substantial meals to each soldier. The standard practice is to provide one hot cooked meal for the other meal whenever possible. A heater or oven is not included since an Esbit cooker is part of each soldier's personal equipment. Enough food items are contained within the Einpersonenpackung to sustain the soldier for 24 hours. Currently there are three menus; each includes two meals out of a selection of 19 meals, with several heavy-duty foil trays containing items such as lentils with sausages, Yugoslav Sausage, Goulash, beef burgers in tomato sauce, Italian pasta, or Tofu stir-fry. There are also three smaller foil "cans" of bread spreads such as cheese spread, liver-sausage, dried-meat sausage, or cheese spread with green peppers. The meal box also includes: thinly sliced rye bread (170 g), hard crackers (1100 kcal), a foil can of fruit salad, instant cream of wheat, instant fruit juice powder, instant coffee, instant tea, powdered cream, a chocolate bar, sugar, salt, gum, jam, water purifying tablets, two plastic bags, matches, paper towels, and a user guide. The Einpersonenpackung ration is supplied in two types, rations 1 to 5 are packaged in a grey cardboard box with the meals packaged in sealed heavy duty foil trays which may be heated by immersing in hot water. The trays are opened using a knife or other sharp implement. Rations 6 to 19 are packaged in a resealable carry pouch, which are either NATO Olive coloured, desert brown or transparent. The meals are packed in retort pouches.
=== Filtration === The Celle engineer, Wilhelm Berkefeld, recognized the ability of diatomaceous earth to filter and developed tubular filters (known as filter candles) fired from diatomaceous earth. During the cholera epidemic in Hamburg in 1892, these Berkefeld filters were used successfully. One form of diatomaceous earth is used as a filter medium, especially for swimming pools. It has a high porosity because it is composed of microscopically small, hollow particles. Diatomaceous earth (sometimes referred to by trademarked brand names such as Celite) is used in chemistry as a filtration aid, to increase flow rate, and filter very fine particles that would otherwise pass through or clog filter paper. It is also used to filter water, particularly in the drinking water treatment process and in fish tanks, and other liquids, such as beer and wine. It can also filter syrups, sugar, and honey without removing or altering their color, taste, or nutritional properties.

As of October 2024, Novo was the second most valuable drug company in the world by market capitalization, second only to its competitor Eli Lilly and Company. By July 2025, Novo had fallen to become the fifth most valuable drug company amid rising competition from generic weight loss substitutes. The company named Maziar Mike Doustdar as the new CEO, effective August 7, 2025. In March 2025, the company announced new plans for a direct-to-consumer offering of its Wegovy weight loss drug. The company established a new pharmacy, called NovoCare, which would charge customers $499 per month for access to the drug, less than half the cost of the drug through other pharmaceutical distribution networks. In September 2025, Novo announced that it would reduce its global workforce by 9,000 people, approximately 9% of its total workforce. In October 2025, Novo acquired Akero Therapeutics for $5.2 billion. Akero Therapeutics is a clinical stage company focusing on the treatment of metabolic dysfunction-associated steatohepatitis (MASH).
== Expression == In placental mammals, including humans, the highest levels of DAO expression are observed in the digestive tract (intestinal mucosa) and the placenta. DAO expression is also observed in kidney of various species. DAO is also expressed in eosinophils.
Robinson, and Scott Sehon wrote about the merits of taking an anti anti-communist position that does not deny the atrocities but make a distinction between anti-authoritarian communist and other socialist currents, both of which have been victims of repression.
Renin (etymology and pronunciation), also known as an angiotensinogenase, is an aspartic protease protein and enzyme secreted by the kidneys that participates in the body's renin–angiotensin–aldosterone system (RAAS), which increases the volume of extracellular fluid (blood plasma, lymph, and interstitial fluid) and causes arterial vasoconstriction. Thus, it increases the body's mean arterial blood pressure. Renin is not commonly referred to as a hormone, although it has a receptor, the (pro)renin receptor, also known as the renin receptor and prorenin receptor (see also below), as well as enzymatic activity with which it hydrolyzes angiotensinogen to angiotensin I.

Reports describe modulation of GABA signalling, changes in monoamine turnover and effects on neurotrophic factor expression. These observations come mainly from animal and cell studies. A single unifying mechanism has not been demonstrated.
The peptide enters plasma rapidly and is broken down by ordinary proteases into amino acids and shorter fragments. Reported half-lives are short. Whether meaningful amounts of the intact molecule reach the brain is an open question.
Most clinical reports are small, published in Russian and not independently replicated. English-language reviews highlight the absence of large randomised trials. Conclusions about efficacy should be treated as provisional.
Intranasal administration predominates in both animal and human research because it avoids hepatic first-pass metabolism. Injectable and intraperitoneal routes appear in animal work mainly for comparison.
