The short version of heptapeptide fits in a sentence. The long version — which is the one that helps — is below.
Reviewed 2026-08-01. Anything still debated is marked as such rather than presented as settled.
Outside its country of origin the compound is generally handled as a research chemical rather than an approved medicine. No regulatory approval from the United States Food and Drug Administration or the European Medicines Agency has been granted for human use. Identity and purity are normally checked by reverse-phase high-performance liquid chromatography, with mass spectrometry used to confirm the molecular mass. Lyophilised material is stored cold and desiccated, and repeated freeze-thaw cycles are avoided.
Published work on this peptide almost always uses intranasal delivery, with drops or a spray applied to the nasal mucosa. Some animal experiments have used subcutaneous or intraperitoneal injection, and a smaller number have compared routes directly. Oral administration is not a focus of the literature, because short peptides of this size are broken down by digestive enzymes and cross intestinal barriers poorly. How much of an intranasal dose reaches the bloodstream intact in humans remains an open question.
Lyophilized Selank, the dry powder form, is generally stored frozen at minus 20 degrees Celsius or colder for long-term keeping. The solid is hygroscopic and should stay sealed, dry, and protected from light. Once dissolved, the peptide is less stable and is usually held refrigerated at 2 to 8 degrees Celsius for short periods. Repeated freezing and thawing is avoided because it can promote aggregation and loss of activity. Buffers and pH choice also affect how long a solution remains usable.
Solubility behavior is a practical concern for handling. Selank dissolves readily in water and in common aqueous buffers, which simplifies preparation of working solutions. The choice of solvent, ionic strength, and pH can influence aggregation over time, particularly at higher concentrations. Aqueous solutions are typically sterile-filtered before use. Because stability depends on several variables, storage and handling notes should be treated as general guidance rather than fixed rules, and specific values are best confirmed against a certificate of analysis for each batch.
Characterization of Selank in a laboratory setting relies on standard peptide methods. Reverse-phase high-performance liquid chromatography separates the target from related impurities and provides a purity figure, commonly reported as 95 percent or higher. Mass spectrometry, typically electrospray ionization or matrix-assisted laser desorption, confirms the molecular mass and helps detect truncation or modification. Amino acid analysis can verify composition when a sequence-level check is needed. These techniques together establish identity and purity for a given lot.

Published clinical evidence is limited. Most controlled trials were conducted in Russia, enrolled modest numbers of participants, and appeared in Russian-language journals, which restricts independent verification. Reported outcomes include lower anxiety scores, improved attention and memory measures, and changes in fatigue ratings. Reviews written in English note methodological limitations such as small samples and inconsistent endpoints. Whether the compound produces clinically meaningful benefit relative to established anxiolytics is therefore an open question rather than an established finding.
Selank is a synthetic heptapeptide with the sequence Thr-Lys-Pro-Arg-Pro-Gly-Pro. It was designed at the Institute of Molecular Genetics of the Russian Academy of Sciences as a structural analogue of tuftsin, a naturally occurring tetrapeptide fragment of the immunoglobulin heavy chain. The added Pro-Gly-Pro tail was intended to slow enzymatic degradation and extend biological activity. In Russia it is registered as an anxiolytic nasal preparation, while regulators elsewhere have not approved it for clinical use.
Development took place at the Institute of Molecular Genetics of the Russian Academy of Sciences, where a series of short peptides were designed in the 1980s and 1990s. Selank was selected from variants of tuftsin that showed resistance to plasma peptidases. Russian regulatory approval covers it as an anxiolytic agent given intranasally. Outside that market the compound is normally handled as a research chemical rather than a medicine, and no widely recognised international pharmacopoeial monograph exists. The name Selank is a coined trade designation rather than a systematic chemical name.
Enzymatic stability motivates the extra three residues at the carboxyl end. Native tuftsin is cleaved quickly by circulating aminopeptidases and carboxypeptidases, which limits its duration of action and its usefulness as a tool compound. Extending the chain with proline-rich segments is a common design tactic because proline constrains the backbone and slows proteolysis. The same Pro-Gly-Pro motif appears in other Russian-developed peptides of the era. Whether the full seven-residue chain is required for activity, or whether it acts mainly as a prodrug releasing tuftsin, remains unresolved.
Selank is a synthetic heptapeptide with the sequence Thr-Lys-Pro-Arg-Pro-Gly-Pro. It was designed as a stabilized analogue of tuftsin, a naturally occurring tetrapeptide fragment derived from the immunoglobulin heavy chain. The additional Pro-Gly-Pro segment at the carboxyl terminus is intended to slow enzymatic cleavage. The compound is usually described in the literature as a synthetic peptide with anxiolytic and cognitive-related activity, a label that reflects a research context rather than an approved therapeutic category.
Most published work on selank originates from a small number of research groups in the Russian Federation. A large share of that record appears in Russian-language journals, which limits access for readers who rely on English-indexed databases. Independent replication by laboratories outside the original research network is sparse in publicly available sources. This concentration of origin and language is a frequently noted feature when the compound is summarized in broader reviews of synthetic peptides.
In 2019, the FDA warned consumers that kratom remains unapproved for interstate commerce for use as a drug, may be unsafe in commercially available products, and is on an import alert, which can lead to confiscation of imported supplies. Efforts to schedule kratom generated significant controversy, both among the general public and the scientific community, and were ultimately unsuccessful. In August 2025, Florida Attorney General James Uthmeier announced an emergency rule placing 7-hydroxymitragynine into Schedule I status under Florida state law, without any mention of a carve out or any exclusions for Mitragyna speciosa which contains low amounts of 7OH, effectively making kratom illegal in Florida. On April 10, 2026, Kansas Governor Laura Kelly signed a bill into law banning the sale and possession of all kratom products in the state, including 7-OH, effective July 1, 2026. North Dakota banned kratom, effective August 5, 2026, via an executive order from the governor and also an emergency rule from the pharmacy board. Subsequently, the legislature passed a law banning synthetic kratom but allowing natural kratom to be sold to those over 21, along with requiring labeling and imposing marketing restrictions. This law was signed by the governor on September 4th, but kratom is still banned until regulations under the new law are issued.
== Enzyme function == Cyanophycinase is a carboxy terminal specific exopeptidase, meaning it catalyzes the cleavage of the carboxy terminal peptide bond of cyanophycin. It was hypothesized that cyanophycinase was a serine protease due to extreme inhibition of the enzyme when used with known serine protease inhibitors, such as DMSO. Site directed mutagenesis experiments confirmed that the enzyme is a serine protease and suggested that Ser 132 is the primary catalytic residue. Other key residues for specificity include Gln101, Asp172, Gln173, Arg178, Arg180 and Arg183 which form a conserved pocket adjacent to Ser 132. Kinetic characterization of the enzyme demonstrates that the enzyme displays Michaelis–Menten kinetics with a kcat of 16.5 s−1 and a kcat/KM of 7.5 × 106 M−1 s−1.
==== American Federation of Government Employees v. Trump ==== A lawsuit was filed on February 6, 2025, by the American Foreign Service Association and the American Federation of Government Employees in the US District Court for the District of Columbia, requesting a temporary restraining order and preliminary injunction against the administration, claiming that it violated separation of powers, the Take Care Clause of the Constitution, and the Administrative Procedure Act and requesting that all attempts to shut down the agency be halted, all recent actions be reversed, and a new acting director be appointed. The following day, U.S. District Judge Carl Nichols, nominated by President Trump in 2019, stated from the bench that he would enter in a "temporary restraining order", pausing the plan to put thousands of employees on leave and pausing the accelerated removal of workers from their posts abroad. On February 21, 2025, Judge Nichols cleared the way for the Trump administration to move forward with pulling thousands of USAID staffers off the job in the United States and around the world, as part of an administration plan to also provide those abroad with a 30-day deadline to move back to the U.S. at government expense. Nichols had previously argued that Trump's actions threaten the safety of USAID workers abroad because many are deployed in unstable regions.
== Function == PBPs are all involved in the final stages of the synthesis of peptidoglycan, which is the major component of bacterial cell walls. Bacterial cell wall synthesis is essential to growth, cell division (thus reproduction) and maintaining the cellular structure in bacteria. Inhibition of PBPs leads to defects in cell wall structure and irregularities in cell shape, for example filamentation, pseudomulticellular forms, lesions leading to spheroplast formation, and eventual cell death and lysis. PBPs have been shown to catalyze a number of reactions involved in the process of synthesizing cross-linked peptidoglycan from lipid intermediates and mediating the removal of D-alanine from the precursor of peptidoglycan. Purified enzymes have been shown to catalyze the following reactions: D-alanine carboxypeptidase, peptidoglycan transpeptidase, and peptidoglycan endopeptidase. In all bacteria that have been studied, enzymes have been shown to catalyze more than one of the above reactions. The enzyme has a penicillin-insensitive transglycosylase N-terminal domain (involved in formation of linear glycan strands) and a penicillin-sensitive transpeptidase C-terminal domain (involved in cross-linking of the peptide subunits) and the serine at the active site is conserved in all members of the PBP family. Some low-molecular-weight PBPs associate with the MreB cytoskeleton and follow its rotation around the cell, inserting petipdoglycan in an oriented manner during cell growth.

According to the Bureau of Transportation Statistics, Nuevo Laredo is the busiest border crossing in terms of truck crossings with over 1.7 million trucks a year, more than double than any other crossing on the Mexico–United States border. Nuevo Laredo is the fourth-busiest border crossing in terms of passenger vehicles. Patrick Corcoran of InSight Crime believes that the turf war in Nuevo Laredo will bring a huge wave of violence, but also mentioned that the circumstances have changed since the split of the Gulf Cartel and Los Zetas in early 2010. The current alliance between Guzmán's Sinaloa Cartel and the Gulf Cartel may successfully extract Los Zetas and give Guzmán the upper hand. Once the Sinaloa Cartel gets established in Nuevo Laredo, it may possibly make moves to control Reynosa and Matamoros, Tamaulipas. In 2018, InSight also stated that infighting had developed with CJNG by March 2017 which resulted in the killing of a cartel financier and the leader of the CJNG's hit squad.
== Synthesis == Neuropeptides are synthesized from inactive precursor proteins called prepropeptides. Prepropeptides contain sequences for a family of distinct peptides and often contain duplicated copies of the same peptides, depending on the organism. In addition to the precursor peptide sequences, prepropeptides also contain a signal peptide, spacer peptides, and cleavage sites. The signal peptide sequence guides the protein to the secretory pathway, starting at the endoplasmic reticulum. The signal peptide sequence is removed in the endoplasmic reticulum, yielding a propeptide. The propeptide travels to the Golgi apparatus where it is proteolytically cleaved and processed into multiple peptides. Peptides are packaged into dense core vesicles, where further cleaving and processing, such as C-terminal amidation, can occur. Dense core vesicles are transported throughout the neuron and can release peptides at the synaptic cleft, cell body, and along the axon. A single animal may use hundreds of different neuropeptides. In C. elegans, for example, 120 genes specify more than 250 neuropeptides.
== Withdrawal == Withdrawal symptoms may occur during gradual or particularly abrupt withdrawal of tricyclic antidepressant drugs. Possible symptoms include: nausea, vomiting, abdominal pain, diarrhea, insomnia, headache, nervousness, anxiety, dizziness and worsening of psychiatric status. Differentiating between the return of the original psychiatric disorder and clomipramine withdrawal symptoms is important. Clomipramine withdrawal can be severe. Withdrawal symptoms can also occur in neonates when clomipramine is used during pregnancy. A major mechanism of withdrawal from tricyclic antidepressants is believed to be due to a rebound effect of excessive cholinergic activity due to neuroadaptations as a result of chronic inhibition of cholinergic receptors by tricyclic antidepressants. Restarting the antidepressant and slow tapering is the treatment of choice for tricyclic antidepressant withdrawal. Some withdrawal symptoms may respond to anticholinergics, such as atropine or benztropine mesylate.
Nalmefene, sold under the brand name Revex among others, is a medication that is used in the treatment of opioid overdose and alcohol dependence. Nalmefene belongs to the class of opioid antagonists and can be taken by mouth, administered by injection, or delivered through nasal administration. In terms of its chemical structure and biological activity, nalmefene is similar to another opioid antagonist called naltrexone, as they are both derivatives of opiates. However, nalmefene offers certain advantages over naltrexone. These include a longer elimination half-life, which means it stays in the body for a longer duration, improved absorption when taken by mouth, and no observed liver toxicity that is dependent on the dosage. Nalmefene is available as a generic medication.

== Nerve supply == Heart rate depends on the rate at which the sinoatrial node produces action potentials. At rest, the human heart rate is between 60 and 100 beats per minute. This is a result of the activity of two sets of nerves, one acting to slow down action potential production (these are parasympathetic nerves) and the other acting to speed up action potential production (sympathetic nerves). Modulation of heart rate by ANS is carried by two types of channel: Kir and HCN (members of the CNG gated channels). The sympathetic nerves begin in the thoracic region of the spinal cord (in particular T1-T4). These nerves release a neurotransmitter called noradrenaline (NA). This binds to a receptor on the SA node membrane, called a beta-1adrenoceptor. Binding of NA to this receptor activates a G-protein (in particular a Gs-Protein, S for stimulatory) which initiates a series of reactions (known as the cAMP pathway) that results in the production of a molecule called cyclic adenosinemonophosphate (cAMP). This cAMP binds to the HCN channel (see above). Binding of cAMP to the HCN increases the flow of Na+ and K+ into the cell, speeding up the pacemaker potential, so producing action potentials at a quicker rate and increasing heart rate. An increase in heart rate is known as positive chronotropy. The parasympathetic nerves supplying the SA node (in particular the Vagus nerves) originate in the brain. These nerves release a neurotransmitter called acetylcholine (ACh). ACh binds to a receptor called an M2 muscarinic receptor, located on the SA node membrane.
===== Sensitivity to airborne moisture ===== MOFs are frequently sensitive to moisture in the air. In particular, IRMOF-1 degrades in the presence of small amounts of water at room temperature. Studies on metal analogues have unraveled the ability of metals other than Zn to stand higher water concentrations at high temperatures. To compensate for this, specially constructed storage containers are required, which can be costly. Strong metal-ligand bonds, such as in metal-imidazolate, -triazolate, and -pyrazolate frameworks, are known to decrease a MOF's sensitivity to air, reducing the expense of storage.
Between 1963 and 1968, three Lodges of the Grand Lodge of Cuba in Exile were founded in Miami, named Liberty, Equality, and Fraternity. As of 2010, the Grand Lodge of Cuba in Exile stated that the Communist government of Cuba maintains Freemasonry as a tool and method for control of the population, rather than allowing it to autonomously uphold the ideals of Freemasonry.
Most of the run-through schemes are in stand-alone specialties (e.g., radiology, public health, histopathology), but there are also a few traditionally surgical specialities which can be entered directly without completing core surgical training (e.g., neurosurgery, obstetrics & gynaecology, ophthalmology). The length of this training varies; for example, general practice is three years while radiology is five years. The UK grade equivalent of a US fellow in medical/surgical sub-specialties is the specialty registrar (ST3–ST9) grade of sub-specialty training. However, while US fellowship programmes are generally 2–3 years in duration after completing the residency, UK trainees spend 4–7 years. This generally includes service provision in the main specialty; this discrepancy lies in the competing demands of NHS service provision, and UK postgraduate training stipulating that even specialist registrars must be able to accommodate the general acute medical take—almost equivalent to what dedicated attending internists perform in the United States (they still remain minimally supervised for these duties). In 2024, the British Medical Association (BMA) advocated for all junior doctors to be renamed residents to prevent the confusion between resident doctors and medical students that terms such as "junior doctors" and "doctors in training" produce.

Most published work uses intranasal application, either as drops or as a nasal spray. Injection routes appear in a smaller set of animal experiments. Oral use is uncommon in the literature because peptide breakdown and poor absorption limit this route.
Reported human trials are few, brief and originate mainly from one country. They describe effects on anxiety-related measures, but sample sizes are small and reporting is limited. The results are widely regarded as preliminary rather than confirmed.
Reverse-phase liquid chromatography is used to assess purity and separation of related impurities. Mass spectrometry confirms that the observed molecular mass matches the expected peptide. A certificate of analysis from the supplier is the usual document of record.
Reverse-phase HPLC is the usual method and gives a percentage purity value. Mass spectrometry then confirms the molecular mass. Together they provide a basic identity and purity profile for a lot.
